Pluripotency Factor-Mediated Expression of the Leptin Receptor Links Obesity to Oncogenesis through Tumor-Initiating Stem Cells
Researchers demonstrate that robust and selective expression of the receptor for the adipocyte-derived peptide hormone leptin is a characteristic feature of tumor-initiating stem cells and of a broad array of embryonic and induced pluripotent stem cells and is mediated directly by the core pluripotency-associated transcription factors OCT4 and SOX2. Activin and BMP4 Synergistically Promote Formation of Definitive Endoderm in Human Embryonic Stem Cells
Scientists show that combining a second related ligand, BMP4, in combination with Activin A yields 15 to 20% more definitive endoderm as compared to Activin A alone. [Stem Cells] Abstract

Thermal Stability of FGF Protein Is a Determinant Factor in Regulating Self-Renewal, Differentiation and Reprogramming in Human Pluripotent Stem Cells
Here researchers show that only a few FGF family members (FGF2, FGF4, FGF6 and FGF9) are able to sustain strong ERK phosphorylation and NANOG expression levels in human ES cells. [Stem Cells] Abstract

Protein Kinase C Mediated Extraembryonic Endoderm Differentiation of Human Embryonic Stem Cells
Here investigators show that phorbol ester 12-O-Tetradecanoylphorbol 13-acetate causes human embryonic stem (ES) cells to undergo an epithelial mesenchymal transition and to differentiate into cells expressing markers of parietal endoderm, another extraembryonic lineage. They further confirmed that this differentiation is through the activation of PKC pathway and demonstrated that a particular PKC subtype, PKC-delta, is most responsible for this transition. Differentiation Efficiency of Induced Pluripotent Stem Cells Depends on the Number of Reprogramming Factors
In order to elucidate the influence of factor reduction on subsequent differentiation, scientists compared the efficiency of neuronal differentiation in induced pluripotent stem cells (iPSC) generated from postnatal murine neural stem cells with either one (Oct4; iPSC1F-NSC), two (Oct4, Klf4; iPSC2F-NSC) or all four factors (iPSC4F-NSC) with those of embryonic stem cells and iPSC produced from fibroblasts with all four factors (iPSC4F-MEF). Yin Yang 1 Extends the Myc-Related Transcription Factors Network in Embryonic Stem Cells
Researchers show that, in mouse embryonic stem cells, Yin Yang 1 has genome-wide Polycomb group protein-independent activities while it is still stably associated with the INO80 chromatin-remodeling complex, as well as with novel RNA helicase activities. [Nucleic Acids Res] Abstract

Acute Depletion of Tet1-Dependent 5-Hydroxymethylcytosine Levels Impairs LIF/Stat3 Signaling and Results in Loss of Embryonic Stem Cell Identity
Here investigators demonstrate that, unlike genetic deletion, RNAi-mediated depletion of Tet1 in mouse embryonic stem cells (mESCs) led to a significant reduction in 5-hydroxymethylcytosine and loss of mESC identity. [Nucleic Acids Res] Abstract
